今天我们来八卦一下RNAi是怎么发现的（老板八卦给我们的）……

 

早在dsRNA可诱导RNAi发现以前，做小虫的就已经开始把antisense strands of mRNA transcripts（类似morpholino）直接注射到小虫的性腺里面，希望虫卵能吸收morpholino，并在小虫的下一代直接knock-down基因表达。

 

后来，Fire实验室来了个“伟大”的graduate student，“错误地”将morpholino注射到了intestine里面而不是gonad里面，结果得到了huge effects。

 

之后，Fire他们就开始把morpholino往intestine里面打。不过不久他们就发现，这个morpholino纯化得越干净，效果越不好。

 

Fire同学phd的时候是做in vitro transcription的。Gut feeling告诉他估计是dsRNA在起作用，于是一个伟大的technique就这样诞生了。

 

只得一提的是，那个错误地将morpholino打到intestine的poor graduate student后来被kicked out of graduate school了。

 

从这个故事我们学到了：we have to make mistakes on purpose. If other people ask why you do it, you can simply tell them this story and say "I wanna win the Nobel Prize."
